Overview

Regulator of G protein signaling 6 (RGS6) is a member of the RGS protein family, which accelerate the deactivation of heterotrimeric G proteins by enhancing the intrinsic GTPase activity of Gα subunits, thereby regulating G protein-coupled receptor (GPCR) signaling[2][4][5][7]. RGS6 is part of the R7 subfamily and contains both DEP (disheveled, EGL-10, pleckstrin homology) and GGL (Gγ subunit-like) domains, which control membrane targeting, stability, and subcellular localization. RGS6 modulates a wide range of cellular processes, including neuronal activity, apoptosis (cell death), mitochondrial signaling, and tumor suppression, primarily through G protein-dependent and independent pathways[2][3][7]. Its dysfunction or altered expression is associated with several diseases, notably various forms of cancer, neuropsychiatric disorders, and alcohol use disorder, making it an emerging but complex therapeutic target[2][3][4][7].
