Overview

Regulator of microtubule dynamics protein 3 (RMDN3), also widely referred to as protein tyrosine phosphatase-interacting protein 51 (PTPIP51), is a microtubule-associated and mitochondrial protein involved in the regulation of calcium homeostasis, cellular differentiation, cytoskeleton organization, apoptosis, and cell proliferation[1][2][6][10]. RMDN3 is primarily located in the mitochondrial outer membrane and cytoplasm, where it interacts with a range of proteins including PTP1B, 14-3-3, Raf-1, and c-Src, linking it to major signaling pathways such as MAPK[2]. Its best-documented biological effect is the induction of apoptosis, suggested to occur via mitochondrial destabilization and cytochrome c release[2][10]. RMDN3 is expressed in a wide spectrum of tissues, especially differentiated cells, and has been clinically associated with various malignancies, notably prostate and skin cancers, as well as certain neurodegenerative diseases[1][2][6]. There is currently no evidence for approved drugs that directly target RMDN3, nor for its established use as a diagnostic or therapeutic biomarker[1][2][5][6][10].
