Overview

Regulator of MON1-CCZ1 complex (RMC1) is a highly conserved protein that functions as a scaffolding subunit within the MON1-CCZ1-RMC1 trimeric complex in metazoans, which is crucial for the activation of the small GTPase RAB7A and thus for endosomal maturation and autophagic flux[1][2]. RMC1 binds both MON1 and CCZ1, facilitating assembly of the complex and distinguishing the metazoan mechanism of endolysosomal trafficking from that in yeast and fungi, which lack this component[1][4]. The WD40 domain of RMC1 enables protein-protein interactions and likely provides a binding platform, while its unique MIC domain is important for complex assembly[4]. RMC1 localizes to endosomes and lysosomes and is important for lysosomal biology, protein trafficking, and possibly organismal development[1][4]. There is evidence implicating RMC1 in cancer, specifically colon cancer, although it is not established as a therapeutic target and no known drugs currently interact with this protein[4].
