Overview

Regulator of ribosome synthesis 1 (RRS1) is a highly conserved nucleolar protein essential for ribosome biogenesis, where it enables binding to 5S rRNA and mediates maturation and assembly of the 60S ribosomal subunit[1][2][3][4]. RRS1 plays a central role in chromosome alignment during mitosis, is involved in cell cycle regulation, and stabilizes nucleolar structure by interacting with other ribosome biogenesis factors[1][3][4]. Aberrant expression of RRS1 has been observed in multiple cancers, including hepatocellular carcinoma and breast cancer, where it facilitates oncogenic transformation by supporting rapid cell proliferation and suppressing p53-mediated growth arrest[1][3][5]. RRS1 dysfunction is also implicated in neurodegeneration and other diseases through its effects on ribosome assembly and cellular homeostasis[1]. No direct drugs targeting RRS1 are currently described in the literature.
