Regulator of thyroid function and cancer (RTFC), encoded by the C14orf93 gene, is a recently characterized human protein implicated in thyroid biology and cancer. RTFC was identified as a susceptibility gene for familial non-medullary thyroid cancer (FNMTC), with several missense mutations (e.g., R115Q, V205M, G209D) shown to enhance thyroid cancer cell survival, migration, and the ability to transform normal thyroid cells[6][2][4][1]. In vitro knockout studies in mouse embryonic stem cells demonstrate that RTFC promotes thyroid lineage differentiation, and loss of function impairs this process[1][2][4]. RTFC is predicted to enable RNA binding activity and may act as a transcriptional regulator involved in positive regulation of gene expression and cell differentiation[2][3][5]. RTFC is also recognized as a potential antigen in certain peripheral T-cell lymphomas[2]. However, no clinically approved drugs are known to target RTFC, and its detailed molecular mechanism and broader physiological roles remain under investigation.
