Overview

Regulatory factor X2 (RFX2) is a member of the regulatory factor X family of transcription factors, characterized by a highly conserved winged helix DNA-binding domain[5][6][7]. It acts as a transcriptional activator, binding DNA as a monomer or as a heterodimer with other RFX family members, and plays critical roles in the regulation of gene expression, particularly genes required for ciliogenesis, motile cilia function, and spermatogenesis[2][3][4][6]. RFX2 is essential for proper formation and function of cilia in multiciliated epithelial cells and for the meiotic and haploid phases of spermatogenesis, with loss of RFX2 resulting in male infertility in mice due to impaired differentiation of germ cells and failure of flagellum formation[2][4]. RFX2 also influences the expression of testis-specific genes and has broader roles in the development, homeostasis, and disease by regulating both ciliary and non-ciliary gene networks[3][6][7]. The gene is structurally related to other RFX family members including RFX1, RFX3, RFX4, and RFX5[5][7]. There are no widely documented drugs or small molecules directly targeting RFX2, reflecting its role as a transcription factor rather than a classic therapeutic target[6][7]. It is not classified as a receptor, enzyme, transporter, or ion channel. There are no known clinical biomarkers or safety concerns specifically associated with RFX2 modulation.
