Overview

Regulatory macrophages (Mregs) are a subset of macrophages with potent immunoregulatory properties. They are characterized by high IL-10 production, variable TGF-β production, and low IL-12 production. Mregs are induced by dual stimulation with a TLR ligand (e.g., LPS) and a secondary signal (e.g., PGE₂). They suppress inflammation, promote tolerance, and can be used therapeutically for autoimmune diseases and transplant rejection, but their immunosuppressive nature presents infection risks.

Mechanism of action

Macrophage polarization towards an anti-inflammatory and tolerogenic phenotype, suppressing pro-inflammatory cytokine production (IL-1β, IL-6, TNFα) and promoting Treg expansion via TGF-β secretion.
