Overview

Regulatory solute carrier protein family 1 member 1 (RSC1A1) is a regulatory protein that modulates the expression and membrane localization of key solute carriers, primarily the sodium/glucose cotransporter SLC5A1 and the organic cation transporter SLC22A2. It exerts its action through transcriptional and post-transcriptional mechanisms, including inhibition of transporter protein exocytosis via a dynamin and PKC-dependent pathway and regulation of vesicle trafficking from the trans-Golgi network. The protein may be found in the trans-Golgi network or in the nucleus depending on the cell cycle stage, and is implicated in glucose-dependent short-term inhibition of transporter activity. Disease associations include lysosomal mannosidosis, though direct roles in common therapeutic indications are not well-established

Mechanism of action

Transcriptional and post-transcriptional inhibition of SLC5A1 and SLC22A2, inhibition of dynamin/PKC-dependent exocytosis, regulation of vesicle release from trans-Golgi network

03

Biological functions

Regulation of sodium/glucose cotransporter SLC5A1 expression and activityRegulation of organic cation transporter SLC22A2 expressionNegative regulation of D-glucose transmembrane transportInhibition of exocytosis of SLC5A1 proteinPossible short-term glucose-dependent regulation of transporter activity
04

Disease associations

Mannosidosis, beta A, lysosomalno widely established roles in cancer or inflammation have been confirmed
05

Safety considerations

Not characterized; as RSC1A1 regulates solute carrier proteins, off-target modulation could disrupt glucose or organic cation homeostasis, but therapeutic safety data is not available
06

Interacting drugs

None directly documented; modulation of transporter activities (SLC5A1, SLC22A2) may indirectly affect drug transport, but no direct pharmacological modulators of RSC1A1 are reported in current literature
07

Biomarkers

None established for patient selection or efficacy monitoring; potential biomarker value in transporter-related pathologies not reported
