Overview

Regulatory subunit of type II PKA R-subunit domain containing 1 (RIIAD1) is a protein-coding gene that encodes a protein characterized by a dimerization/docking (D/D) domain similar to that found in the RII subunits of cAMP-dependent protein kinase A (PKA)[1][3][6]. RIIAD1 belongs to the RIIa clan of D/D domain-containing proteins and is closely related to ROPN1, ROPN1L, SPA17, and CATIP as part of the R2D2 structural subgroup[2]. The canonical feature of this protein is to interact with A-kinase anchoring proteins (AKAPs), which compartmentalize PKA signaling within cellular microdomains[2]. Although structural studies support its role in protein-protein interactions, RIIAD1 itself is not an enzyme, receptor, or traditional signaling molecule and is not currently identified as a direct therapeutic target, nor are there known small-molecule drugs or therapeutic antibodies that target it[1][3][5][6]. Its biological importance appears to relate to scaffolding and potentially in structural organization of flagellar and ciliary architecture, with implications for cell signaling localization and possibly motility, but no specific endemic roles in cancer or other diseases have been established[2].
