Overview

Regulatory T cells (Tregs) are a specialized subpopulation of CD4+ T lymphocytes that play a central role in maintaining immune homeostasis, self-tolerance, and preventing autoimmune disease. They achieve this by suppressing the activation and proliferation of other immune cells, particularly effector T cells. Tregs employ multiple mechanisms to exert their immunosuppressive effects, including cell-contact dependent suppression, cytokine-mediated suppression (IL-10, TGF-β), and metabolic disruption. Targeting Treg activation is being explored for both autoimmune disease treatment (enhancing function) and cancer immunotherapy (inhibiting/regulating function).
