Overview

Regulatory T cell (Treg)-mediated immunosuppression refers to the collection of mechanisms by which regulatory T cells suppress immune responses, particularly those of effector T cells and other immune cell types. This process is essential for maintaining self-tolerance, preventing autoimmune disease, and modulating immune responses during infection, inflammation, and cancer. Tregs employ multiple overlapping mechanisms including secretion of inhibitory cytokines (IL-10, TGF-β, IL-35), cytolysis (granzyme B), metabolic disruption (IL-2 consumption, adenosine generation), modulation via inhibitory receptors (CTLA-4, PD-1), and direct cell contact.
