Overview

Regulatory T lymphocytes (Tregs) are a specialized subset of CD4+ T lymphocytes that play a central role in maintaining immune tolerance and homeostasis within the intestinal environment. They suppress effector immune responses against self-antigens, food antigens, and commensal bacteria, preventing autoimmunity or chronic inflammation. Intestinal Treg populations arise from thymus-derived (tTregs) and peripherally induced (pTregs) sources, with pTregs differentiating in the gut due to factors like retinoic acid, TGF-β, and signals from commensal microbiota. Defective function or numbers of intestinal Tregs contribute to inflammatory bowel diseases, making them a therapeutic target for autoimmune/inflammatory disorders affecting the gastrointestinal tract.
