Overview

Replication factor C subunit 2 (RFC2) is a 40 kDa protein that forms part of the heteropentameric replication factor C (RFC) complex, also known as activator 1, which is essential for the elongation of primed DNA templates during DNA replication and repair[1][2][4][5]. RFC2 binds ATP and is critical for loading the proliferating cell nuclear antigen (PCNA) onto DNA[1][2][4]. Disruption of RFC2 has been linked to Williams syndrome[1][2][5]. Dysregulation of RFC2 is associated with malignant progression in various cancers, where it serves as a prognostic biomarker and influences tumor mutational burden, microsatellite instability, and immune infiltration[2][3]. RFC2 interacts with proteins including PCNA, BRD4, CHTF18, RFC4, and RFC5, but it is not currently targeted by any approved drugs[1][2][4].
