Replication protein A 14 kDa subunit (RPA3) is one of three subunits forming the heterotrimeric replication protein A (RPA) complex, the major single-stranded DNA-binding complex in eukaryotes[1][2][3][7]. RPA3 is important for stabilizing single-stranded DNA intermediates during essential processes such as DNA replication, repair, and recombination. By binding to and protecting single-stranded DNA, the RPA complex prevents secondary structure formation and recruits multiple repair factors and DNA damage response proteins to chromatin[1][2][4][7]. RPA3, in particular, is thought to play a role in the polarity of DNA binding within the complex and is essential for maintaining genome integrity[1][4][7]. Defects or dysregulation of RPA3 have been associated with cancer and inherited syndromes such as Bloom Syndrome[1][6].
