Overview

Replication protein A 30 kDa subunit (RPA4) is a component of the replication protein A (RPA) complex, a heterotrimeric single-stranded DNA-binding factor essential for eukaryotic DNA metabolism[1][2][3][4][5]. RPA4 substitutes for the canonical RPA2 subunit in certain contexts and participates in DNA replication, repair (notably nucleotide excision repair), homologous recombination, and cell cycle checkpoint activation[1][4][5]. It binds single-stranded DNA to prevent secondary structure formation and protect DNA during processing, and is involved in assembling or recruiting repair and replication proteins to sites of DNA damage or replication stress[1][4][5]. RPA4 has distinct expression patterns and appears important in specific tissues (e.g., colon, placenta)[4], localizes to DNA repair foci, and may contribute to cellular DNA damage responses and inhibition of viral replication[5]. Its role in cancer and response to genotoxic stress is an area of ongoing research.
