Replikin peptide motifs are specific amino acid sequences found in pathogen proteins that are characterized by a high concentration of lysine and histidine residues. These motifs are defined by having at least two lysines separated by 7 to 10 amino acids, at least one histidine, and a total lysine content of 6% or more (Bogoch & Bogoch, 2004, Proteome Science). Research suggests that the concentration of these motifs, known as the Replikin count, correlates with the rate of replication in viruses and bacteria, such as influenza and SARS-CoV-2 (Bogoch & Bogoch, 2005, Nature Precedings). In the context of drug development, Replikins are targeted as conserved epitopes for synthetic vaccines and monoclonal antibodies. By targeting these motifs, therapeutic interventions aim to disrupt the pathogen's ability to replicate rapidly and provide broad-spectrum protection against evolving strains. The most prominent drug candidate associated with this target is Panreuvax, a synthetic vaccine designed to provide immunity against multiple influenza strains by targeting shared Replikin sequences. Despite these applications, the concept remains specialized and has faced scrutiny regarding its predictive universality in mainstream virology.
