The M2-1 protein is a critical regulatory protein encoded by the human respiratory syncytial virus (HRSV). It acts as a transcriptional processivity and antitermination factor essential for viral gene expression. It prevents premature termination during transcription by binding competitively to both viral RNA and the phosphoprotein (P). It also enhances readthrough of intergenic junctions during viral mRNA synthesis, allowing production of full-length or polycistronic RNAs. Due to its essential role in RSV replication/transcription—and absence from host cells—M2–1 represents an attractive antiviral drug target.
