REST Corepressor 1 (CoREST) is a transcriptional corepressor that plays a critical role in neural cell differentiation. In complex with REST, CoREST recruits chromatin-modifying enzymes, inducing chromatin condensation and transcriptional repression. It is essential for regulating neurogenesis by repressing neuronal gene expression in non-neuronal cells during development. Mutations or dysregulation involving RCOR1 have been associated with diseases including Joubert Syndrome 1 and medulloblastoma.
