REST corepressor 2 (RCOR2), also known as CoREST2, is a protein encoded by the RCOR2 gene in humans. It is a 58 kDa, 523-amino acid nuclear protein that functions as a transcriptional corepressor, particularly in the nervous system. RCOR2 is a member of the CoREST protein family and partners with the RE1-silencing transcription factor (REST) to repress neuronal gene expression in non-neuronal cells, primarily by recruiting chromatin-modifying enzymes to establish a repressive chromatin state. Unlike other members of the CoREST family, RCOR2 can also repress gene expression independently of REST and is thought to regulate unique sets of genes involved in neurodevelopment. RCOR2-mediated complexes play a key role in balancing neural progenitor proliferation and differentiation during brain development. Its physiological functions rely on protein–protein interactions and the recruitment of histone deacetylases and demethylases, rather than direct DNA binding
