REST corepressor 3 is a chromatin-associated protein that functions as a transcriptional corepressor together with REST and complementary chromatin-modifying enzymes, including histone deacetylases and lysine-specific demethylases. It is mainly located in the cytosol and nucleoplasm and modulates gene expression through chromatin condensation, resulting in the suppression of neuronal and other cell-type-specific genes. RCOR3 can operate both in REST-dependent and REST-independent mechanisms, suggesting independent regulatory roles in neurodevelopment and disease processes. Its expression and activity have been linked to disease prognosis in hepatitis B and have potential relevance for other disorders characterized by aberrant gene repression.
