Overview

Resting lymphocyte kinase (RLK), also known as TXK, is a member of the Tec family of non-receptor tyrosine kinases. It is primarily expressed in T-cells and natural killer (NK) cells, where it plays a critical role in intracellular signaling pathways that regulate immune cell development, function, and differentiation. RLK facilitates signal transduction from surface receptors such as the T-cell receptor (TCR) and Fc receptor and is involved in activating downstream signaling molecules including phospholipase Cγ1 (PLCγ1), leading to activation of nuclear factor of activated T-cells (NFAT), nuclear factor κB (NFκB), and mitogen-activated protein kinase (MAPK) pathways. RLK is an attractive target for therapeutic intervention in certain T-cell malignancies, as well as inflammatory or autoimmune diseases.
