Reticular cells are specialized stromal cells of mesenchymal origin that provide the structural framework for lymphoid organs, including the lymph nodes, spleen, and bone marrow (StatPearls, 2023). They synthesize reticular fibers, primarily composed of type III collagen, which form a complex meshwork that supports the organization and movement of lymphocytes and other immune cells (Wikipedia, 2024). Beyond their mechanical role, fibroblastic reticular cells (FRCs) are essential for immune homeostasis, as they secrete key chemokines such as CCL19 and CCL21 to facilitate the interaction between T cells and dendritic cells (Nature Reviews Immunology, 2013). These cells also form conduits that allow for the rapid transport of small molecules and antigens from the lymph to the high endothelial venules (PubMed, 2015). In pathological conditions, reticular cells are involved in the development of lymphoid fibrosis and can be co-opted by malignant cells to promote tumor survival and metastasis (Journal of Clinical Investigation, 2016). They play a role in chronic inflammatory diseases where they can undergo remodeling that impairs normal immune function (Frontiers in Immunology, 2020). While they are critical components of the immune microenvironment, they are considered a cell type rather than a discrete molecular therapeutic target. Consequently, there are no drugs that specifically target reticular cells as a primary mechanism of action, though their signaling pathways are areas of active research.
