Overview

The Reticuloendotheliosis virus (REV) gp90 protein is the surface (SU) subunit of the viral envelope glycoprotein complex, playing a critical role in the initial stages of viral infection. It is responsible for recognizing and binding to specific host cell receptors, such as the inorganic phosphate transporter PiT-1 (SLC20A1), which facilitates viral entry into avian and occasionally mammalian cells. Following receptor binding, gp90 works in tandem with the transmembrane (TM) subunit, often designated gp20, to mediate fusion between the viral envelope and the host cell membrane. In veterinary medicine, gp90 is a primary target for the development of subunit vaccines and diagnostic assays aimed at controlling REV outbreaks in poultry, which cause significant economic losses through runting-stunting syndrome and various lymphoid tumors. While no small-molecule pharmacological agents are currently approved for targeting gp90 in humans, it remains a focal point for research in avian health and the development of retroviral-based gene therapy vectors. Understanding the structural dynamics of gp90 is essential for the design of targeted entry inhibitors and more effective neutralizing antibodies.
