Reticulon-1 is an integral membrane protein most commonly localized to the tubular regions of the endoplasmic reticulum, where it helps generate and stabilize the tubular ER network. It contains a conserved Reticulon Homology Domain (RHD) with two long hydrophobic segments, and can oligomerize. Reticulon-1 regulates key cellular functions such as ER organization, nuclear envelope formation, calcium homeostasis, apoptosis, and autophagy. Dysregulation of reticulon proteins, in particular RTN-1C, has been implicated in the development of cancer and a range of neurodegenerative diseases, where it may serve as a biomarker or therapeutic target. Reticulon-1 can modulate enzymes like HDACs and PDIs, affecting processes like protein folding and aggregation in neurodegeneration.
