Reticulon 2 is one of four mammalian reticulon proteins characterized by a conserved carboxy-terminal reticulon homology domain (RHD) with two unusually long hydrophobic membrane-associated domains flanking a central hydrophilic loop[1][2][3]. It localizes mainly to the endoplasmic reticulum, where it is thought to play a major role in shaping ER membrane curvature and maintaining ER structure[2]. RTN2 influences protein processing, secretion, and possibly vesicle transport between ER and other intracellular compartments[2][3]. Recent research demonstrates it is upregulated in gastric cancer, enhancing the migration, invasion, and metastatic behavior of cancer cells, likely through the activation of ERK signaling and promotion of epithelial-to-mesenchymal transition (EMT)[3]. Reticulons overall are also implicated in neurodegenerative disease pathways, highlighting their diverse cellular roles[1][3].
