Reticulon-4 receptor-like 1 (RTN4RL1) is a cell surface receptor predominantly expressed in the nervous system, where it plays a functionally redundant role in postnatal brain development, axon regeneration, and formation of the corpus callosum. It restricts axon outgrowth and regeneration by interacting with extracellular chondroitin sulfate proteoglycans and signaling via Rho activation, influencing the actin cytoskeleton. RTN4RL1 protects motoneurons from apoptosis and is also implicated in the regulation of dendritic spine and synapse number during brain development. Diseases associated with RTN4RL1 include Miller-Dieker lissencephaly syndrome and microcephaly. While no current drugs specifically target RTN4RL1, agents affecting the Rho/ROCK pathway, which is downstream of Nogo receptor signaling (NgR1-3), are in clinical trials for CNS injury
