Reticulophagy regulator family member 3 (RETREG3) is an endoplasmic reticulum-anchored protein that functions as an autophagy regulator, predominantly in neurons[1][2]. It localizes to ER tubules and sheet edges, where it interacts with curvature-inducing proteins like RTN4B to modulate ER morphology. RETREG3 is involved in reticulophagy, a process facilitating ER turnover under conditions of cellular stress, promoting ER fragmentation and subsequent autophagic clearance[2]. Its expression is crucial for neuronal differentiation, with roles in neurite outgrowth and axon morphogenesis. Loss of RETREG3 impairs axonal growth and dendrite formation. RETREG3 acts in a protein-containing complex and is found in various neural tissues as well as other organs[1][8][11].
