Retinal dehydrogenase and other zinc-dependent visual enzymes represent a functional group of proteins essential for ocular health and the visual cycle. The primary enzyme in this category is a zinc-dependent alcohol dehydrogenase (ADH) that facilitates the oxidation of retinol (Vitamin A) to retinal, the immediate precursor for the light-sensitive pigment rhodopsin (Vallee & Bazzone, 1983; Duester, 1996). Other enzymes in this group include superoxide dismutase (SOD), which protects the retina from oxidative stress, and carbonic anhydrase, which is involved in fluid dynamics and pH regulation within the eye (Ugarte & Osborne, 2001). Zinc serves as a critical catalytic or structural cofactor for these enzymes, and its deficiency is a well-documented cause of night blindness (nyctalopia) and a risk factor for the progression of age-related macular degeneration (AMD) (AREDS Research Group, 2001). Pharmacological management typically involves zinc supplementation to maintain enzyme activity and support the structural integrity of the retinal pigment epithelium. Conversely, these enzymes can be inhibited by drugs like disulfiram or competed for by ethanol, which may interfere with normal visual function and retinoid signaling (Amory et al., 2011).
Zinc acts as a required catalytic cofactor for alcohol dehydrogenase (functioning as a retinol dehydrogenase) to convert retinol to retinal, and as a structural or catalytic component of antioxidant enzymes like superoxide dismutase to protect retinal tissues.
