The retinal membrane phospholipids and macular pigment compartment represent a specialized physiological region in the macula responsible for protecting the eye from oxidative stress and maintaining visual function. The macular pigment consists of three xanthophyll carotenoids—lutein, zeaxanthin, and meso-zeaxanthin—which are concentrated in the Henle fiber layer and inner plexiform layer (Bernstein et al., 2016; PMID: 26541886). These pigments serve as a biological filter for high-energy blue light and act as antioxidants to quench singlet oxygen and free radicals generated during the visual cycle. Retinal membrane phospholipids, particularly those enriched with docosahexaenoic acid (DHA), are vital for the structural integrity and signaling kinetics of photoreceptor outer segments (Bazan, 2006; PMID: 16431876). A reduction in the density of these pigments or the integrity of these lipids is a significant risk factor for the development and progression of age-related macular degeneration (AMD). Therapeutic strategies, such as those evaluated in the AREDS and AREDS2 clinical trials, focus on dietary supplementation to replenish these components and mitigate retinal damage (AREDS2 Research Group, 2013; PMID: 23644932).
