Overview

Retinal pigment epithelial (RPE) cell loss refers to the reduction in number or function of RPE cells, a monolayer of pigmented cells located between the neural retina and the choroid. These cells are essential for retinal health, supporting photoreceptor survival, phagocytosing shed photoreceptor outer segments (POS), maintaining the blood-retina barrier, and regulating nutrient/waste exchange. Loss or dysfunction of RPE cells is implicated in several degenerative retinal diseases, most notably age-related macular degeneration (AMD), leading to secondary death of overlying photoreceptors and vision loss. Key mechanisms contributing to RPE cell loss include aging, oxidative stress, inflammation, and genetic factors.
