Retinoic acid receptor beta (RAR-beta) is a member of the nuclear hormone receptor superfamily that functions as a ligand-activated transcription factor [6, 10]. It plays a critical role in mediating the effects of retinoic acid, the active metabolite of vitamin A, on essential biological processes such as cell growth, differentiation, and apoptosis [2, 13]. RAR-beta typically forms heterodimers with retinoid X receptors (RXRs) to bind specific DNA sequences known as retinoic acid response elements (RAREs), thereby regulating the expression of target genes involved in development and homeostasis [6, 10]. In many cancers, RAR-beta acts as a tumor suppressor, and its expression is frequently lost or silenced through epigenetic mechanisms like promoter hypermethylation [11, 12]. Drugs like adapalene, a third-generation synthetic retinoid, selectively target RAR-beta and RAR-gamma to treat dermatological conditions like acne by modulating skin cell turnover and reducing inflammation [1, 5, 17]. Beyond dermatology, RAR-beta is being investigated for its roles in neurogenesis and as a potential therapeutic target in various malignancies [3, 19].
