Overview

Retinoic acid-related orphan nuclear receptor gamma t (RORγt) is a specific isoform of the RAR-related orphan receptor gamma (RORγ), a nuclear receptor transcription factor. RORγt, encoded by the *RORC* gene, is a master regulator of immune cell development, especially in T helper 17 (Th17) cells. It's crucial for thymocyte survival and Th17 differentiation, activating genes for IL-17A, IL-17F, and other pro-inflammatory cytokines. RORγt maintains Th17 cell identity and influences circadian rhythms. As Th17-mediated inflammation is linked to autoimmune diseases, RORγt is an attractive therapeutic target. Inhibition aims to alleviate autoimmune symptoms without broad immunosuppression. However, RORγt also plays roles in cancer and has context-dependent functions, impacting both pro-inflammatory and immunosuppressive responses.
