Overview

Retinoic acid-related orphan receptor gamma t (RORγt) is a member of the nuclear receptor superfamily and a ligand-dependent transcription factor. It is a key regulator of Th17 cell differentiation and function, driving the expression of pro-inflammatory cytokines. RORγt is implicated in various autoimmune diseases and potentially in cancer. It is considered a druggable target with both orthosteric and allosteric ligand binding sites. The RORC gene encodes two isoforms: RORγ and RORγt, with RORγt being selectively expressed in immune tissues.

Mechanism of action

Binds specific DNA response elements as a monomer or dimer to activate or repress gene transcription, depending on ligand binding status and cellular context. Ligand binding induces conformational changes affecting coactivator/corepressor recruitment at target gene promoters/enhancers.
