Retinol dehydrogenase 11 (RDH11) is an enzyme primarily involved in the metabolism of retinoids, catalyzing the oxidation and reduction of retinol (vitamin A alcohol) and retinaldehyde (vitamin A aldehyde) using NADP(H) as a cofactor. RDH11 shows high activity towards 9-cis, 11-cis, and all-trans forms of retinol and is a member of the short-chain dehydrogenase/reductase (SDR) family. It is expressed notably in the human prostate and other tissues, and its expression can be regulated by androgens. Mutations in the RDH11 gene have been linked to retinal dystrophy, juvenile cataracts, and short stature syndrome. RDH11 operates mainly in the endoplasmic reticulum membrane and plays a role in the visual cycle and the detoxification of aldehydes. No clinically approved drugs directly target RDH11, and it is not currently used as a biomarker for patient selection or therapeutic efficacy.
