Retrograde Golgi transport protein RGP1 homolog (RGP1) is a regulatory subunit of the Ric1-Rgp1 complex, which acts as a guanine nucleotide exchange factor (GEF) specifically for the Rab6A GTPase, a master regulator of Golgi and post-Golgi trafficking. Rab6A, when activated (GTP-bound), is necessary for proper vesicular transport, Golgi apparatus organization, and retrograde trafficking of select receptors (such as mannose 6-phosphate receptors) from endosomes to the Golgi. Ric1 and Rgp1 must act together to facilitate the nucleotide exchange on Rab6A, promoting the active GTP-bound form necessary for these trafficking events. The Ric1-Rgp1 complex structure is distinct from other Rab GEFs, with Ric1 containing β-propeller and α-solenoid domains, and Rgp1 containing an arrestin-like domain critical for Golgi localization and interaction with Rab6A.
