Overview

Retrotransposon Gag-like protein 8C (RTL8C) is a small, truncated protein derived from an ancestral retrotransposon gag gene and is a member of the Mart/RTL8 family of domesticated retroelement proteins[1]. RTL8C is highly homologous to PEG10 in the N-terminal capsid-like region but lacks a C-terminal domain typical of canonical retroelement Gag proteins. RTL8C interacts directly with PEG10, leading to decreased formation and extracellular release of PEG10-derived virus-like particles, possibly by interfering with capsid assembly. RTL8C also forms a tripartite regulatory node with PEG10 and the ubiquitin-binding protein UBQLN2, where RTL8C is required for UBQLN2-mediated degradation of PEG10. Although essential in PEG10 regulation and cellular stress granule dynamics, there is currently no evidence that RTL8C serves directly as a therapeutic target, nor are there known drugs or biomarker uses associated with this molecule[1][2][3].
