01

Overview

The RevCAR peptide epitope is a synthetic short peptide domain incorporated into the extracellular portion of engineered T cells (or NK cells) in the RevCAR system. This peptide serves as a unique, orthogonal recognition site for adapter molecules known as RevCAR target modules (RevTMs), which are bispecific antibodies that simultaneously bind this synthetic epitope and a tumor-associated antigen (e.g., fibroblast growth factor-inducible 14 or PD-L1). This system allows researchers and clinicians to control activation of the engineered immune cells in a stimulus-dependent manner, enhancing therapeutic safety and modularity. The peptide’s function is not derived from endogenous biology but is a tool for precise immune cell redirection in adoptive cell therapy platforms[3][5][7].

Mechanism of action

Enables switchable, reversible activation of engineered immune cells when used with bispecific RevCAR target modules (RevTMs)

Biological functions

Immune cell retargeting (as part of a synthetic therapeutic system)Modular binding ligand for bispecific adapter molecules

Safety considerations

Off-target immune activation possible if peptides are mis-expressed or immune system is improperly stimulated via adapterThe system is designed to improve CAR T-cell safety by allowing “on/off” control via the peptide epitope–adapter interaction
