Overview

RGS5 antisense RNA 1 (RGS5-AS1), also known as lnc-RGS5, is a long non-coding RNA (lncRNA) that overlaps with the protein-coding gene RGS5, but does not encode a protein itself. RGS5-AS1 has been identified as a novel lncRNA (LOC127814295, ENSG00000232995) that is deregulated in various cancers, especially breast cancer, where its higher expression is associated with enhanced tumor cell proliferation and reduced overall survival. Mechanistically, RGS5-AS1 is thought to act as a competing endogenous RNA by sponging miR-542-5p, thereby upregulating critical oncogenic factors like FoxM1 and the VEGFA/Neuropilin 1 pathway, contributing to tumor growth and progression. There are no specific therapeutic drugs targeting RGS5-AS1 at this time, but it may have potential as a biomarker or therapeutic entry point in certain cancers[3].
