Overview

Endogenous ammonia transporters, primarily the Rhesus (Rh) glycoproteins RhBG and RhCG, are specialized membrane proteins that facilitate the movement of ammonia (NH3) and ammonium (NH4+) across biological membranes (Weiner & Verlander, 2017). These transporters are critical for maintaining systemic acid-base balance and nitrogen homeostasis, particularly in the kidneys and liver (Handlogten et al., 2005). In the renal system, RhCG is the primary pathway for ammonia secretion into the collecting duct, which is essential for the excretion of metabolic acids (Biver et al., 2008). Dysregulation or genetic mutations in these transporters are associated with clinical conditions such as distal renal tubular acidosis and hyperammonemia (UniProt, 2024). While they are vital for physiological function, there are currently no FDA-approved drugs that directly target Rh glycoproteins, though they remain a subject of research for treating metabolic and hepatic disorders (PubMed, 2023). Their role in the ammonia shuttle makes them a potential target for future therapeutic interventions in chronic kidney disease and liver failure (NIH, 2024).
