Rho-associated coiled-coil containing protein kinases (ROCKs) are serine/threonine kinases that play a central role in regulating the actin cytoskeleton, cell shape, and movement. They are major downstream effectors of the small GTPase RhoA and belong to the AGC family of protein kinases. There are two main isoforms in mammals: ROCK1 and ROCK2. Dysregulation or abnormal activation of ROCK has been implicated in several diseases, making it an attractive therapeutic target.
