Overview

Rho family GTPase 2 (RND2) is a small (~21 kDa) signaling G protein and a Rho family small GTPase encoded by the RND2 gene in humans. Members of the Rho GTPase family are key regulators of actin cytoskeleton organization in response to extracellular growth factors, and RND2 is specifically involved in the regulation of neuronal morphology, endosomal trafficking, and smooth muscle contractility. RND2 is unique within its family as it promotes RhoA signaling, in contrast to other Rnd proteins that antagonize RhoA signaling, impacting cytoskeletal dynamics and neurite outgrowth in neuronal cells. It is expressed in neuronal tissues (e.g., cortical and hippocampal regions), in neuron-like PC12 cells, and in uterine myometrial tissues. Mutations or dysfunction in RND2 are linked to rare developmental syndromes but no drugs currently target this protein. RND2’s main functions are intracellular signaling and cytoskeletal modulation rather than acting as an extracellular receptor or conventional drug target.
