Overview

Rho GDP dissociation inhibitor alpha (ARHGDIA) is a cytosolic regulator that maintains members of the Rho GTPase family—including CDC42, RAC1, and RHOA—in their inactive, GDP-bound states by preventing GDP dissociation and subsequent GTP binding. ARHGDIA extracts Rho GTPases from membranes into the cytosol, regulates their stability, protects them from degradation, and contributes to the proper distribution and recycling of the active and inactive forms. Through modulation of Rho GTPase signaling, ARHGDIA plays an essential role in cell motility, migration, cytoskeletal dynamics, and cellular homeostasis. Mutations or changes in ARHGDIA expression are associated with hereditary nephrotic syndromes and cancer cell migration and invasion.
