Overview

Rho GDP dissociation inhibitor gamma (ARHGDIG) is a cytosolic regulatory protein that inhibits the release of GDP from Rho family GTPases, thereby maintaining them in their inactive (GDP-bound) state[1][2][3]. It is highly expressed in the brain, kidney, and pancreas and shows distinct features, including a unique N-terminal domain likely important for subcellular localization[1]. ARHGDIG is essential in neural stem cell biology, where it maintains the undifferentiated state and restricts migration. It also modulates the activity of Rho GTPases such as RhoA, RhoC, and Rac1, impacting cytoskeletal organization and cell motility. In oncology, overexpression of ARHGDIG is correlated with increased tumor stage and metastasis in cancers like lung cancer, suggesting a role in regulating invasive behavior[1]. Genetic variants in ARHGDIG have been linked to certain forms of autosomal recessive deafness and pathologic nystagmus[2]. There are no approved drugs that specifically target ARHGDIG directly, but it is a relevant node in Rho GTPase signaling pathways frequently implicated in cancer and cell migration[2][3][5].
