Rho GTPase-activating protein 10 (ARHGAP10) is an enzyme that regulates signaling cascades by promoting the hydrolysis of GTP bound to Rho family GTPases, converting them from their active (GTP-bound) to inactive (GDP-bound) states. This action suppresses Rho GTPase-mediated processes, influencing actin cytoskeleton dynamics, cell adhesion, and migration. ARHGAP10 contains multiple domains, including BAR, PH, RhoGAP, and SH3, enabling it to perform modular regulatory functions. The gene exhibits alternative splicing, producing variants with context-specific effects. Reduced ARHGAP10 expression has been observed in breast tumor tissue and is associated with advanced cancer stage and poor prognosis, suggesting a tumor suppressor role. Additionally, it participates in apoptosis regulation, and genetic variants are linked to schizophrenia and other neuropsychiatric conditions.
