Overview

Rho GTPase-activating protein 20 (ARHGAP20) is a member of the Rho GTPase-activating protein family, a subclass of enzymes that downregulate Rho family GTPases to control cytoskeletal dynamics, cell motility, and signaling pathways affecting cell adhesion and migration[1][2][3]. ARHGAP20 contains both a RhoGAP domain, which catalyzes the inactivation of Rho GTPases, and an RA domain, potentially acting as an effector for Ras- or Ral-like GTPases[3]. It is ubiquitously expressed, most notably in the brain and testis. Functional evidence demonstrates that ARHGAP20 suppresses cancer cell migration and invasion, at least in part by negatively regulating the PI3K-AKT signaling pathway. Its expression is observed to be altered in multiple cancer types – acting as a tumor suppressor in hepatocellular carcinoma and being upregulated in certain chronic lymphocytic leukemia subtypes with specific chromosomal deletions[1][3]. Decreased ARHGAP20 expression in tumors tends to correlate with greater malignancy and poorer prognosis. As of now, ARHGAP20 is not directly targeted by therapeutics but may serve as a prognostic biomarker or a node for intervention through modulation of associated signaling pathways.
