Overview

Rho GTPase-activating protein 23 (ARHGAP23) is an intracellular enzyme that functions as a GTPase activator for members of the Rho family of small GTPases, notably including RhoA and Cdc42. By stimulating the intrinsic GTPase activity of these small G-proteins, ARHGAP23 inactivates them, thus downregulating signaling pathways that control cytoskeletal organization, cell morphology, and migration. ARHGAP23 contains conserved protein domains—PDZ, pleckstrin homology, and RhoGAP—that are important for substrate recognition and protein-protein interactions. Its activity is important for organizing the actomyosin network and confining cell protrusions, with functional implications in tissue development, neuronal signaling, and cancer progression
