Overview

Rho GTPase activating protein 28 (ARHGAP28) is an enzyme belonging to the RhoGAP family that functions to inactivate Rho-type GTPases via promotion of GTP hydrolysis. It serves as a negative regulator of Rho GTPase signaling, playing a pivotal role in the control of actin cytoskeleton dynamics, stress fiber disassembly, and cell adhesion. ARHGAP28 is found at cell junctions and in the nucleoplasm. It has been implicated in tumor suppression in various experimental models, and expression changes in ARHGAP28 are linked to cancer biology, particularly in meningioma and renal cell carcinoma, as well as to allergic diseases and neurobehavioral traits. Genetic and epigenetic alterations of ARHGAP28 may influence disease progression and therapeutic response, notably affecting sensitivity to drugs such as tyrosine kinase inhibitors in cancer. It also plays a role in tissue regeneration and vascular biology, with potential involvement in migraine pathogenesis[1][3][5][6].
