Rho guanine nucleotide exchange factor 10-like protein (ARHGEF10L) is a guanine nucleotide exchange factor (GEF) belonging to the RhoGEF subfamily. It specifically activates Rho GTPases such as RhoA, RHOB, and RHOC, which are pivotal regulators of the cytoskeleton, cell migration, and many aspects of cell signaling. ARHGEF10L promotes cancer cell proliferation, migration, and epithelial-to-mesenchymal transition via the RhoA–ROCK1–phospho-ERM pathway and is overexpressed in a variety of malignancies, including cervical, liver, and gastric cancers. It is associated with increased tumor aggressiveness and risk, making it a candidate oncogene and a potential molecular marker in tumor biology. The gene has also been implicated in non-cancerous diseases such as cerebral visual impairment. Its functional domains include a Dbl homology domain and a WD40-like domain, and it is mostly cytosolic in distribution[1][2][3].
