Rho guanine nucleotide exchange factor 19 (ARHGEF19) is a member of the RhoGEF (guanine nucleotide exchange factor) family, acting downstream of diverse receptors to regulate Rho GTPase activity—especially RhoA—by facilitating GDP-GTP exchange. It includes typical Dbl homology (DH) and pleckstrin homology (PH) domains. Acting as a signal transduction intermediary, ARHGEF19 regulates cytoskeletal dynamics, cell proliferation, cell migration, and invasion. Evidence shows it is upregulated in certain cancers, including non-small cell lung cancer, where it helps drive tumorigenesis, invasion, and metastasis in part through interaction with BRAF and MAPK pathway activation, independent of canonical RhoA functions. High expression in tumors is correlated with adverse clinical outcomes, suggesting a role as both a poor prognostic factor and a potential, though as yet undeveloped, therapeutic target. No direct pharmacological modulators or clinically actionable drugs are currently known for ARHGEF19. The molecule’s role in MAPK signaling and cancer progression makes it of interest for further therapeutic exploration. Regulatory mechanisms and domain structure are conserved within the Ephexin family, but the full biological diversity and context-specific functions of ARHGEF19, particularly beyond cancer, remain the subject of ongoing research.
