Rho guanine nucleotide exchange factor 37 (ARHGEF37) is a protein-coding gene that encodes a guanine nucleotide exchange factor (GEF), which activates Rho family small GTPases by promoting the exchange of GDP for GTP[1][10]. ARHGEF37 is predicted to be active in the cytoplasm and is implicated in signal transduction pathways, including clathrin-mediated endocytosis, where it assists dynamin 2 at late endocytic sites[6][8]. Genetic variants in ARHGEF37 have been associated with rare diseases such as epidermolysis bullosa simplex 1A and thiamine-responsive megaloblastic anemia syndrome[1]. No drugs are currently known to target ARHGEF37 directly, nor are there established biomarker or safety concerns for therapeutic intervention involving this gene.
